Never spend time socializing with someone when there are customers in line. Someone is going to get peeved enough to report you to management if you do.
If something happens that could hold the line up and you can't deal with it very fast, call a manager to deal with it so you don't hold the line up.
Even when you are tired, even when you are 8.5 hours into what was supposed to be an 8 hour shift on Thanksgiving day and your family is waiting for you, NEVER glare at a customer, give a customer a dirty look, or otherwise show the customer you are not happy to see them. They will most assuredly report you.
There are con artists who will know you are new, and who will do things that could get you fired. If anyone gives you a bill larger than a $5, don't put the bill in the register until you have counted out their change and they have accepted it. (I got hit that way: guy in my line hands me a $10 in payment for something that cost less than $2. I put the bill in the tray, gave the guy his change, closed the register. Guy looks me in the eye and tells me, ';I gave you a $20.'; People in line behind the guy. I couldn't remember whether the guy gave me a $10 or $20. Gave him the extra $10. Came up $10 short. Got docked for it. Lived and learned.)

A lot of this depends on what type of store you work at. But general tips include making eye contact with a customer, greeting them, a friendly or neutral expression. You will find that if you smile, people will treat you better than if you do not. Take the time to learn the inventory that your store carries, even if it means walking around on your off time... some stores are better at training than others. Trust me, customers will be happier having to wait for a price check if you know where to call than if you don't. Take the time to get to know associates on the sales floor. They can alert you when there are good sales and if you are on good terms with them, they are more likely to get an answer for you faster. I know there were cashiers that I liked better than others when I worked retail... and I did work a little faster for those I liked. Not intentional, just what occurred.
Ask other cashiers you work with for tips regarding the specific register. I worked as a cashier for Kmart for awhile and you had to press a button to suspend a transaction to keep the timer from counting and of course nobody said anything about using this while a customer say dug in their wallet for spare change... and it affected my metrics until I learned how to play the game. I was not any slower than the other cashiers initially, but my metrics made me look worse... and once I learned that trick, the actual customer's experience was no faster, but my times improved dramatically.

Greet the customer. Don't take rude customers personal. If you need help with an irate customer, be sure to get a manager. Let them handle the problem if you can not resolve it. Pay attention the cash you are given. Some people may try to argue that they gave you more so they will get more change back. Make sure to wish them a good day/night at the end of transaction.
I think it is always nice when the cashier says ';hi, how are you today?'; and/or thank you. it makes shopping a much more pleasant experience that just being ingored when you check out.
Be friendly. It's really annoying to have a cashier who is rude and unfriendly.
Also don't let asshole customers get to you
Be cautious and will have a sound presents of mind,be cordial to the customers,clients etc
